Description du contenu de l'enseignement

This UE is intended to train CPIAM students in NMR and mass spectrometry instrumentation.
This UE will be articulated around integrated courses and practical sessions on NMR and MS instruments applied to the characterization of chemical and biological molecules.


After four hours of TD dedicated to the bases of NMR and mass spectrometry, the practical content of this UE will be based on the implementation of four days of TP divided in two TP on NMR and two TP on GC-MS and LC-MS. These analytical methods will be used for the analysis of molecules in various domains (chemistry, biology) in order to illustrate the physical mechanisms of these instrumentation and their performances. The students will be divided into small groups. Collaboration with Bruker Daltonics company will allowed students to spend a day in the Bruker Factory in Wissembourg first, to have a vision of NMR and MS markets and second, to see and to understand the conception of NMR and FT-MS superconductor magnet.



Le contenu de l'enseignement correspond à quatre séances de travaux pratiques basées sur l'utilisation et la compréhension instrumental de la RMN, la chromatographie gazeuse couplée à la spectrométrie de masse (GC-MS) et la chromatographie liquide couplée à la spectrométrie de masse (LC-MS).
  • Partie LC-MS et GC-MS:
-Analyse d'un mélange de composés contenant du brome ou du chlore, notion de profil isotopique
-Suivi de réaction de l'aldéhyde cinnamique par GC-MS dans différents solvants
-Dosage du limonène dans une huile essentielle par GC-MS
-Analyse protéomique de la myoglobine de coeur de cheval par LC-MS/MS
  • Partie RMN:
-Présentation du spectromètre (aimant, console, poste de travail)
-Démonstration: expérience 1D 1H
-Applications: LOCK, SHIMS, accord de sonde, choix du solvant
-Préparation d’un échantillon en salle de TP (menthol + 600 uL de CDCl3, env. 10 mmol)
-Application: réalisation d’une expérience 1D 1H, 13C, 13C découplé 1H et 1D 1H sélective. -Introduction à la RMN 2D
-Démonstration: expérience 2D COSY 1H
-Application: expérience 2D COSY 1H, 2D HSQC 1H-13C -Application au choix: DOSY 1H ou NOESY 1H

  • To understand the role of the different components of NMR and MS
  • To practice NMR and MS instrumentation
  • To develop an autonomy on NMR and MS instrumentation
  • To develop an analytical strategy in MS and NMR.
  • To have a first experience in proteomic analysis
